Kualitas Spematozoa Semen Sapi Angus (Bos Taurus) dalam Pengencer Tris Kuning Telur dengan Substitusi Susu Kacang Kedelai

Penelitian ini betujuan untuk mengetahui kualitas spermatozoa sapi Angus (Bos taurus) terhadap motilitas, viabilitas, dan abnormalitas dalam pengencer tris kuning telur (TKT) dengan substitusi susu kacang kedelai (SKD) pada konsentrasi Semen segar ditampung dari sapi Angus umur 4 tahun menggunakan metode vagina buatan, semen dievaluasi secara makroskopis terhadap volume, warna, konsistensi, dan pH dan mikroskopis terhadap gerakan massa, motilitas, viabilitas, abormalitas, dan konsentrasi. Motilitas spermatozoa semen segar >70% diencerkan ke dalam 4 perlakuan P0: TKT (3 mL), P1: TKT (2 mL) + SKD (1 mL), P2: TKT (1,5 mL) + SKD (1.5 mL), P3: TKT (1 mL) + SKD (2 mL) dan P4: TKT (0 mL) + SKD (3 mL). Semen dievaluasi setelah pengenceran kemudian semen disimpan di dalam lemari pendingin dengan suhu 20 °C, evaluasi dilakukan setiap 24 jam. Data hasil penelitian dianalisis berupa motilitas, viabilitas, dan abnormalitas spermatozoa dianalisis menggunakan analisis of variance. Hasil penelitian menunjukan spermatozoa sapi Angus (Bos taurus) tertinggi terdapat nilai rerata persentase motilitas spermatozoa tertinggi terdapat pada P2 dengan nilai (42.98±16.62%), nilai rerata viabilitas spermatozoa tertinggi terdapat pada P2 (58.67±14.16%), dan nilai rerata abnormalitas tertinggi terdapat pada P2 (7.34±0.37%). Berdasarkan hasil TKT dan SKD yang digunakan dapat mempertahankan konfigurasi normal bentuk spermatozoa sehingga dapat meminimalisir persentase abnormalitas. Spermatozoa tidak dapat bertahan lama karena semakin tinggi konsentrasi pengencer yang digunakan maka semakin menurun kualitas spermatozoa.
